I've got two favorites that fit in this category - Knight's Gambit, a book of stories by Faulkner, and Transposed Heads by Thomas Mann. Both old favorites.

The Girlhood of Shakespeare's Heroines. John Crowley. It was published in 2005 so I don't know if it's been long enough to consider it overlooked but it's already out of print and selling for twice the original asking price which says to me that unless one is already a fan or unless it's anthologized somewhere very prominent it isn't likely to be read by many people.
